Linezolid

Linezolid is an antibiotic of the protein synthesis inhibitor class. It kills bacteria by inhibiting binding to the 50s subunit of the ribosome and terminating protein synthesis. It is effective against gram positive strains only.

Linezolid is indicated for the treatment of the following infections caused by susceptible species of bacteria-

  • Community acquire pneumonia
  • Nosocomial pneumonia
  • Skin and skin structure infections

The usual dosage us 600mg twice daily for up to 10-14 days. It is available as tablets, powder for suspension and intravenous infusion.

How to reconstitute suspension from powder

 

Linezolid can cause some GI disorders like nausea, vomiting, abdominal pain & diarrhea. Myelosuppression has also been reported. Overgrowth of Candida sp. can also occur.

  • Do not take Linezolid if you are allergic to linezolid or any ingredients contained in this drug.
  • Patients should be continuously monitored for signs and symptoms of serotonin syndrome, such as cognitive impairment, hyperpyrexia, hyperreflexia, and incoordination, when Linezolid and concomitant serotonergic medications are therapeutically suitable.
  • If signs or symptoms appear, doctors should consider stopping one or both of the drugs.

Contraindication

  • Linezolid formulations are contraindicated for using in patients who have known hypersensitivity to Linezolid or any of the other product components.
  • Linezolid should not be used in patients taking any medicinal product which inhibits monoamine oxidases A or B (e.g. Phenelzine, Isocarboxazid) or within two weeks of taking any such medicinal product. 
  • Patients taking directly and indirectly acting sympathomimetic agents, vasopressive agents (e.g. Epinephrine, Norepinephrine), dopaminergic agents (e.g. Dopamine, Dobutamine), serotonin re-uptake inhibitors, tricyclic antidepressants, serotonin 5-HT1 receptor agonists (triptans), meperidine or buspirone should not take linezolid.

There is no known contraindications of linezolid in terms of food and drinks

  •  Linezolid should not be administered to patients with uncontrolled hypertension, pheochromocytoma, thyrotoxicosis, carcinoid syndrome and/or patients taking directly and indirectly acting sympathomimetic agents
