Lubiprostone
Lubiprostone is a locally acting chloride channel activator that increases chloride-rich intestinal fluid secretion without affecting serum sodium and potassium levels. Lubiprostone works by specifically activating ClC-2, a normal constituent of the human intestine's apical membrane, in a protein kinase A-independent manner. Lubiprostone increases motility in the intestine by increasing intestinal fluid secretion, allowing stool to pass more easily and alleviating symptoms associated with chronic idiopathic constipation.
Lubiprostone is indicated for-
- Treatment of chronic idiopathic constipation in adults
- Treatment of irritable bowel syndrome with constipation in women ≥ 18 years old
Chronic idiopathic constipation
- 24 microgram taken twice daily orally with food and water
- Reduce the dosage in patients with moderate and severe hepatic impairment
Irritable bowel syndrome with constipation
- 8 mcg taken twice daily orally with food and water
- Reduce the dosage in patients with severe hepatic impairment
Most common adverse reactions associated with use of Lubiprostone in chronic idiopathic constipation are-
- nausea
- diarrhea
- headache
- abdominal pain
- abdominal distension
- flatulence
Most common adverse reactions in irritable bowel syndrome with constipation are-
- nausea
- diarrhea
- abdominal pain
- Patients may experience nausea; administering food concurrently may alleviate this symptom.
- Do not give to patients who have severe diarrhea.
- Lubiprostone patients may experience dyspnea within an hour of their first dose. This symptom usually goes away after 3 hours, but it can come back if you take it again.
- Before starting Lubiprostone treatment, evaluate patients who have symptoms of mechanical gastrointestinal obstruction.
Contraindication
Contraindicated in patients hypersensitive to lubiprostone or any component of the medication.
