Nitazoxanide

Nitazoxanide is an antiprotozoal and anthelmintic drug classified as a thiazolide. Its mechanism of action involves inhibiting the pyruvate:ferredoxin oxidoreductase enzyme, which leads to the disruption of the energy metabolism of certain protozoa and helminths. Nitazoxanide has broad-spectrum activity against both protozoa and helminths.

 

Nitazoxanide is indicated for the treatment of diarrhea caused by Cryptosporidium parvum or Giardia lamblia in patients 12 years of age and older. It is also indicated for the treatment of helminthic infections, including Enterobius vermicularis (pinworm), Ascaris lumbricoides (roundworm), and Trichuris trichiura (whipworm), in patients 2 years of age and older.

 

Indicataion Age Dosage Duration
Treatment of diarrhea caused by Giardia lamblia or Cryptosporidium parvum. 1-3 years  5 mL of for Oral Suspension (100 mg nitazoxanide) every 12 hours with food   3 days
4-11 years  10 mL of Oral Suspension (200 mg nitazoxanide) every 12 hours with food
≥12 years 1 Tablet (500 mg nitazoxanide) every 12 hours with food or 25 mL of for Oral Suspension (500 mg nitazoxanide) every 12 hours with food

Common side effects of nitazoxanide include:

  • Abdominal pain
  • Headache
  • Nausea
  • Diarrhea
  • Vomiting
  • Dizziness
  • Flatulence

 

  • Nitazoxanide should be used with caution in patients with hepatic impairment, as it may increase the risk of hepatotoxicity.
  • It should also be used with caution in patients with a history of seizure disorders, as it may lower the seizure threshold.
  • Nitazoxanide should be used during pregnancy only if the potential benefit justifies the potential risk to the fetus.
  • It is not known whether nitazoxanide is excreted in human milk, and caution should be exercised when administering nitazoxanide to nursing mothers.
  • Tablets and Oral Suspension should be taken with food.

Contraindication

Nitazoxanide is contraindicated in patients hypersensitive to any component of the product.
