Nitrazepam

Nitrazepam is a GABA-A receptor agonist. GABA receptors when stimulated produces inhibitory effects on other neuronal signals. Nitrazepam is used as a hypnotic agent.

Nitrazepam is indicated in the following conditions-

  • Insomnia

Adults: 5 mg before retiring. This dose may, if necessary, be increased to 10 mg.

 

Side effects are not very common with the use of nitrazepam. The following side effects have been reported-

  • Drowsiness
  • Rash
  • Urticaria
  • Dizziness
  • Headache
  • Syncope
  • Lethargy
  • Nausea
  • Doses may need to be reduced in patients with chronic pulmonary insufficiency, as well as those with chronic renal or hepatic illness. Patients with severe hepatic impairment should avoid benzodiazepines.
  • Nitrazepam should not be used alone to treat depression or anxiety associated with depression because it increases the risk of suicide in these patients. In patients who have a history of alcohol or drug abuse, benzodiazepines should be taken with extreme caution. Benzodiazepines should not be used as a first-line treatment for psychotic disorder.
  • Concomitant use of Nitrazepam and opioids may result in sedation, respiratory depression, coma and death. Because of these risks, concomitant prescribing of sedative medicines such as benzodiazepines or related drugs such as Nitrazepam with opioids should be reserved for patients for whom alternative treatment options are not possible. If a decision is made to prescribe Nitrazepam concomitantly with opioids, the lowest effective dose should be used, and the duration of treatment should be as short as possible.
  • Use of benzodiazepines may lead to the development of physical and psychological dependence upon these products. The risk of dependence increases when high doses are used, especially when given over long periods.

Contraindication

Contraindicated in patients with known hypersensitivity to the drug or other benzodiazepines, such as-
