Pramipexol
Pramipexole is a non-ergoline dopamine agonist that is used to treat Parkinson's disease and restless legs syndrome (RLS). It stimulates dopamine receptors in the brain, leading to an increase in dopamine levels, which improves motor function and reduces symptoms of RLS.
Pramipexole is used to treat the following conditions:
-
Parkinson's disease: Pramipexole is used to treat the symptoms of Parkinson's disease, including tremors, stiffness, and slow movement.
-
Restless legs syndrome (RLS): Pramipexole is used to treat the symptoms of RLS, including unpleasant sensations in the legs that occur at rest, especially at night.
The dosage of Pramipexole depends on the individual's condition and response to treatment. The medication is typically started at a low dose and gradually increased until the desired effect is achieved. The following are general guidelines for dosing:
-
Parkinson's disease: The recommended starting dose is 0.125 mg three times per day. The dose may be increased every 5-7 days until a therapeutic response is achieved, up to a maximum dose of 4.5 mg per day.
-
Restless legs syndrome (RLS): The recommended starting dose is 0.125 mg once daily, taken 2-3 hours before bedtime. The dose may be increased every 4-7 days until a therapeutic response is achieved, up to a maximum dose of 0.5 mg per day.
The most common adverse reactions associated with Pramipexole use include:
- Nausea
- Dizziness
- Somnolence (drowsiness)
- Fatigue
- Insomnia
- Constipation
- Headache
Pramipexole should be used with caution in patients with the following conditions:
-
Renal impairment: Pramipexole is eliminated from the body by the kidneys, so patients with renal impairment may require a lower dose of the medication.
-
Psychiatric disorders: Pramipexole has been associated with an increased risk of impulse control disorders, such as gambling and compulsive shopping.
-
Hypotension: Pramipexole can cause a drop in blood pressure, especially upon standing, so patients should be monitored for signs of hypotension.
-
Impaired cognitive function: Pramipexole can cause somnolence and fatigue, which may impair cognitive function and the ability to perform tasks that require mental alertness.
Contraindication
Pramipexole is contraindicated in patients with a known hypersensitivity to the medication or any of its components.
