Prochlorperazine Maleate

Prochlorperazine Maleate is a dopamine antagonist, which means it blocks the action of the neurotransmitter dopamine in the brain. Specifically, it blocks the action of dopamine at D2 receptors in the brain, which are found in the hypothalamus, pituitary gland, and the chemoreceptor trigger zone (CTZ) of the brainstem.

 

 

Prochlorperazine Maleate is a medication used to treat symptoms of schizophrenia, severe nausea and vomiting, and anxiety. It is available in various forms, including tablets, suppositories, and injectable solutions.

 

The recommended dosage are as follows-

  • Schizophrenia: The usual adult dose is 10 mg three times daily.
  • Severe nausea and vomiting: The usual adult dose is 10 mg every 6 hours as needed.
  • Anxiety: The usual adult dose is 5 mg three times daily.

 

Common side effects of Prochlorperazine Maleate include-

  • drowsiness

  • constipation
  • amenorrhea
  • blurred vision
  • skin reactions 
  • hypotension 

 

  • Tardive dyskinesia: Prochlorperazine Maleate can cause a rare but serious condition called tardive dyskinesia, which is characterized by uncontrollable movements of the face, tongue, or other body parts. Symptoms of tardive dyskinesia may not appear until after many months or years of treatment. If you develop these symptoms, contact your doctor immediately.

  • Neuroleptic malignant syndrome (NMS): Prochlorperazine Maleate can cause a rare but potentially fatal condition called neuroleptic malignant syndrome (NMS), which is characterized by fever, muscle stiffness, and changes in mental status. If you develop these symptoms, contact your doctor immediately.

  • Drowsiness and dizziness: Prochlorperazine Maleate may cause drowsiness and dizziness. Do not drive or operate heavy machinery until you know how this medication affects you.

  • Blood disorders: Prochlorperazine Maleate may cause blood disorders, such as agranulocytosis (reduced white blood cell counts) or leukopenia (reduced white blood cell counts). If you develop signs of infection, such as fever or sore throat, contact your doctor immediately.

  • Anticholinergic effects: Prochlorperazine Maleate may cause anticholinergic effects, such as dry mouth, constipation, and blurred vision.

  • Pregnancy and breastfeeding: Prochlorperazine Maleate should be used during pregnancy only if the potential benefit justifies the potential risk to the fetus. It is not known if Prochlorperazine Maleate passes into breast milk. Consult your doctor before breastfeeding.

 

Contraindication

Do not use in patients with known hypersensitivity to phenothiazines. 

 

  • Do not use in comatose states or in the presence of large amounts of central nervous system depressants (alcohol, barbiturates, narcotics, etc.).
  • Do not use in pediatric surgery.
  • Do not use in pediatric patients under 2 years of age or under 20 lbs.
  • Do not use in children for conditions for which dosage has not been established.
