Pefloxacin

Pefloxacin is an antibiotic of the fluoroquinolone class. It kills bacteria by inhibiting DNA synthesis. It is a batericidal antibiotic.

Pefloxacin is indicated for the following infections caused by susceptible bacteria:

  • Gastrointestinal infection
  • Urinary tract infection
  • Respiratory tract infection
  • Ear, nose and throat infection
  • Bone and joint infection
  • Skin and soft tissue infection
  • Pelvic infection
  • Abdominal and hepatobiliary infection
  • Septicemia
  • Gonorrhoea
  • Endocarditis
  • Meningitis.
  • Pefloxacin is available as tablets.
  • The usual dosage of pefloxacin is 400 mg 2 times per day. The usual treatment for normal infection lasts for about 7 days.
  • The daily doses should be readjusted in individuals with significant hepatic insufficiency or a decreased blood supply to the liver by increasing the intervals between doses. Renal insufficiency does not necessitate a reduction in dosage.

The common side effects caused by pefloxacin include-

  • Nausea and other gastrointestinal disturbances
  • Headache & Dizziness
  • Joint pain
  • Skin rashes
  • Long QT syndrome
  • It can also halt bone and cartilage growth adolescents.
  • Fluoroquinolones, including Pefloxacin, are associated with an increased risk of tendinitis and tendon rupture in all ages. This adverse reaction most frequently involves the Achilles tendon, and rupture of the Achilles tendon may require surgical repair.
  • Patients using Pefloxacin should be encouraged to drink plenty of water. 
  • In individuals with suspected or established CNS problems, such as epilepsy, or other variables that predispose to seizures and convulsions, it should be administered with caution. 
  • Avoid in patients with known QT prolongation, hypokalemia.

Contraindication

Pefloxacin is contraindicated in patients with known hypersensitivity to Quinolone antibiotics such as-

Pefloxacin should not be taken with milk, mineral supplements or antacids.

Gemifloxacin is contraindicated in-

  • Pregnancy & Lactation
  • Children under 18
  • Tendonitis
